Ephesians 1:18

The eyes of your understanding being enlightened; that ye may know what is the hope of his calling, and what the riches of the glory of his inheritance in the saints,

Regarding spiritual knowledge, no person can understand it apart from the Spirit of God. 1 Cor 2:10-14 "The man without the Spirit does not accept the things that come from the Spirit of God, for they are foolishness to him, and he cannot understand them, because they are spiritually discerned." (E.g. how many times has Larry King heard the message, but he still doesn't get it?)
Prov 14:6 "The mocker seeks wisdom and finds none, but knowledge comes easily to the discerning."
One of the most significant understandings we can get is to see how the root of so much of our struggles or problems is a spiritual problem. We try to solve our problems with worldly answers, but if it's a spiritual problem, it needs to be solved with a spiritual answer. And it is the Holy Spirit that helps us see both the real problem and the real answers. John 16:13 "But when he, the Spirit of truth, comes, he will guide you into all truth."
It's difficult for the Spirit to teach and guide you unless you are giving Him opportunity, by coming to God in prayer and in reading His word. Unless you have the lines of commuinication open (both ways), and are paying attention. 1 Thess 5:19 "Do not quench the Spirit." Does God have to get your attention with a 2x4, or can he get is with a whisper?
"The hope of His calling" - Rom 8:29-30 He foreknew us, then we are predestined, called; justified; and glorified. This is our hope, what we look foreward to. This is our state, but because of that, we look forward to our glorious inheritance.
We are to know our hope and our future glory.
